Many owners treat the puppy vaccination course as a series of appointments to keep without grasping the logic behind it, and the question that comes up most often in the consulting room is why three or four doses are needed when an adult dog only needs one every three years. The answer has nothing to do with vaccine strength, it has to do with the antibodies the puppy received from its mother. Those antibodies protect it in the first weeks, then decline, and while declining they pass through an interval in which they are no longer enough to stop the virus but still enough to neutralise the vaccine. Nobody knows exactly when that happens in an individual puppy, and the whole calendar exists because of that uncertainty.
Maternal antibodies: protection and obstacle at once
Puppies and kittens are born with a small share of antibodies transferred across the placenta and receive the bulk of them from colostrum, the first milk, which they can only absorb during the first hours of life. After that the gut closes that route, and whatever was received is all they can count on until their own immune system starts working.
That endowment is not the same for everyone. It depends on how well immunised the mother was, on how much colostrum each newborn took and on how quickly: in a large litter the one that suckled least starts with lower protection. Maternal antibodies are then consumed at a fairly regular rate, halving every nine to ten days for parvovirus, but starting from different levels they reach zero at different moments.
The window of susceptibility, the real problem to cover
Between the moment maternal antibodies drop below the level that protects against disease and the moment they drop below the level that neutralises the vaccine there is a gap. During that period the puppy is vulnerable to natural infection while still failing to respond to vaccination. This is the window of susceptibility, and it is why most parvovirus cases occur in puppies that had already received one or two doses.

This is where the whole architecture of the primary course comes from: nobody knows which dose will be the effective one, so several are scheduled close together so that at least one lands at the right moment. Core components in dogs are distemper, adenovirus and parvovirus, with rabies added where it is compulsory or where the disease is present. In cats they are panleukopenia, herpesvirus and calicivirus. For kittens, feline leukaemia deserves a separate note: it is recommended during the first year of life, after testing to confirm the kitten is not already infected, because susceptibility is higher at that age and the future lifestyle is not yet settled.
Why the final dose cannot come before sixteen weeks
This is the most misunderstood point of the calendar. Many owners consider the course finished at the third dose, which often falls around twelve weeks. In reality the dose that counts is the one given once maternal antibodies have declined in all or nearly all puppies, and that happens around sixteen weeks.

- Rabies vaccination is given from twelve weeks of age: before that it is not considered valid for movement purposes.
- For travel within the European Union validity starts twenty one days after the first vaccination, so the practical minimum age for travel is around fifteen weeks.

Perfect calendars are rare. Intercurrent illness, late adoptions, puppies found on the street and incomplete records are the norm, and each situation has a rule of thumb.
